产品介绍This gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This nuclear enzyme is involved in processes such as chromosome condensation, chromatid separation, and the relief of torsional stress that occurs during DNA transcription and replication. It catalyzes the transient breaking and rejoining of two strands of duplex DNA which allows the strands to pass through one another, thus altering the topology of DNA. Two forms of this enzyme exist as likely products of a gene duplication event. The gene encoding this form, alpha, is localized to chromosome 17 and the beta gene is localized to chromosome 3. The gene encoding this enzyme functions as the target for several anticancer agents and a variety of mutations in this gene have been associated with the development of drug resistance. Reduced activity of this enzyme may also play a role in ataxia-telangiectasia. [provided by RefSeq, Jul 2010]

TROP2（Tumor-Associated Calcium Signal Transducer 2）是一种跨膜糖蛋白，通过激活钙信号通路、MAPK/ERK通路和PI3K/AKT通路，调控肿瘤细胞增殖、侵袭和转移。在三阴性乳腺癌、结直肠癌、胃癌、肺癌等多种实体瘤中高表达，而在正常组织中低表达，是理想的肿瘤诊断标志物和治疗靶点。

实验操作关键要点

IHC-P优化：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复15分钟，可增强胞膜信号；

阳性对照：推荐使用三阴性乳腺癌组织切片（肿瘤细胞胞膜呈强阳性）；

封闭处理：5% BSA室温封闭30分钟，降低非特异性结合。

WB注意事项：

样本处理：肿瘤细胞需用含1% Triton X-100的RIPA裂解液冰上裂解30分钟；

抗体稀释：一抗用5%脱脂奶粉+TBST按1:800稀释，4℃孵育过夜，HRP标记二抗（1:5000）室温孵育1小时。

临床应用与研究价值

靶向治疗：TROP2单抗是ADC药物的核心组件， 用于治疗转移性三阴性乳腺癌，通过将拓扑异构酶I抑制剂SN-38靶向递送至肿瘤细胞，临床响应率达33.3%；

诊断应用：可通过IHC检测肿瘤组织中TROP2表达水平，指导ADC药物用药选择；
