Actins are highly conserved proteins that are involved in various types of cell motility and are ubiquitously expressed in all eukaryotic cells.In vertebrates 3 main groups of actin isoforms, alpha, beta and gamma have been identified. The alpha actins are found in muscle tissues and are a major constituent of the contractile apparatus. The beta and gamma actins coexist in most cell types as components of the cytoskeleton and as mediators of internal cell motility. Actin proteins have very high homology.

The immunogen for this product is based on the ACTC1 protein but this antibody will also recognize: ACTB, ACTA1, ACTG1, ACTA2, ACTG2.

靶点与功能

细胞角蛋白（Cytokeratins，CKs）是上皮细胞 intermediate filament 的主要成分，由54个成员组成（酸性CK9-20和碱性CK1-8）。广谱PCK抗体通常识别CK5/6、CK8、CK18、CK19等多种亚型，核心应用价值包括：

肿瘤来源鉴别：95%以上上皮源性肿瘤（如肺癌、乳腺癌、肠癌）阳性，而间叶组织肿瘤（肉瘤）、淋巴瘤阴性，是判断“癌vs肉瘤”的**标志物；

转移灶溯源：对不明原发灶转移癌（CUP），PCK阳性提示上皮来源，结合CK7/CK20谱可缩小原发部位范围（如CK7+/CK20-提示肺/乳腺来源）；

微小病灶检测：在淋巴结微转移灶（<2mm）检测中灵敏度达90%，显著高于HE染色（65%）。

实验操作关键要点

IHC-P病理诊断（转移癌样本）：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（石蜡切片），上皮细胞胞质信号显著增强；

阳性定位：上皮细胞胞质呈细丝状或弥漫性棕黄色阳性，鳞癌呈网状分布，腺癌呈细颗粒状；

对照设置：必须包含皮肤组织（阳性对照，鳞状上皮强阳性）和平滑肌瘤（阴性对照，间叶组织阴性）。

WB检测（细胞系验证）：

样本选择：HepG2（肝癌，表达CK8/18）、A549（肺癌，表达CK5/18）作为阳性对照，HT1080（纤维肉瘤，阴性对照）；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜250mA恒流1.5小时（0.45μm PVDF膜）；

抗体孵育：AE1/AE3推荐1:200稀释（5%脱脂奶粉+TBST），4℃孵育过夜，曝光可见45-68kDa多条特征条带。

FC检测循环肿瘤细胞（CTC）：

与CD45抗体联合使用（PCK+/CD45-定义为CTC），抗体1μg/1×10⁶细胞，4℃避光孵育30分钟，流式缓冲液洗涤后上机。
