产品介绍This protein is a member of the paired box (PAX) family of transcription factors, typically containing a paired box domain, an octapeptide, and a paired-type homeodomain. This family plays critical roles during fetal development and cancer growth. The specific function of the PAX8 is unknown but it may involve kidney cell differentiation, thyroid development, or thyroid dysgenesis. Alternative splicing in the gene by inclusion or exclusion of exons 7 and/or 8 has produced several known products but the biological significance of the variants is unknown. Several other splice variants have been proposed but the full nature of these products has not been described. Pax8 is also a marker of otic progenitor cells.

实验操作关键要点

WB内参标准化（细胞裂解液）：

上样量控制：推荐20-30μg总蛋白/泳道（PCNA在多数细胞系中表达稳定，CV<8%）；

抗体稀释：单抗1:2000稀释（5%脱脂奶粉+TBST），室温孵育1小时（无需4℃过夜，节省时间）；

定量分析：使用ImageJ软件计算PCNA条带灰度值，目的蛋白灰度值需除以PCNA灰度值校正上样误差。

IHC-P增殖活性评估：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复15分钟（石蜡切片），核信号显著增强；

阳性判读：细胞核呈棕黄色颗粒状阳性，计数5个高倍视野（×400）的阳性细胞比例，PCNA index=（阳性细胞数/总细胞数）×100%；

内参适用性：在细胞周期同步化实验中，PCNA表达随S期比例升高而增加，需结合β-actin等组成型内参综合判断。
