靶点与功能

TSPAN8（Tetraspanin 8）属于四跨膜蛋白超家族（TM4SF），通过四个跨膜结构域锚定细胞膜，参与细胞黏附、迁移、信号转导及外泌体介导的细胞间通讯。在结直肠癌、胰腺癌、卵巢癌等恶性肿瘤中高表达，通过调控整合素（如α3β1）和生长因子受体（如EGFR）活性促进肿瘤侵袭转移，也是外泌体标志物之一，可用于肿瘤微环境互作研究。

实验应用与操作建议

肿瘤转移机制研究：

IHC-P样本：推荐使用结直肠癌或胰腺癌组织芯片，抗原修复采用柠檬酸钠缓冲液（pH6.0）微波修复15分钟，可观察到TSPAN8在肿瘤细胞膜及侵袭前沿的特异性表达；

功能验证：可联合整合素β1抗体进行双标IF，检测两者共定位情况（激发波长分别选488nm/555nm）。

外泌体鉴定：

提取细胞上清外泌体后，WB检测时建议使用RIPA裂解液（含1% Triton X-100），一抗稀释比例1:800，搭配HRP标记二抗（1:5000），可与CD63、CD81等经典外泌体标志物共同验证。
