产品介绍TM4SF3 is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. TM4SF3 is known to complex with integrins. Its gene is expressed in different carcinomas. The use of alternate polyadenylation sites has been found for TM4SF3 gene.

靶点与功能

TTF-1（NKX2-1）是一种含同源框结构的转录因子，主要调控甲状腺滤泡细胞、肺Ⅱ型肺泡细胞及Clara细胞的发育与分化。在肿瘤诊断中，它是肺腺癌（非黏液性/黏液性）、小细胞肺癌、甲状腺乳头状癌/滤泡状癌的特异性标志物，通过识别肿瘤细胞中的TTF-1蛋白，辅助判断肿瘤组织来源（肺/甲状腺vs其他部位）。

实验操作关键要点

IHC-P优化（肺/甲状腺肿瘤诊断）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波加热至95℃，持续20分钟（石蜡切片），可增强核信号；

阳性对照：甲状腺组织（滤泡细胞核阳性）或肺腺癌组织（Ⅱ型肺泡细胞核阳性）；

显色判断：细胞核呈棕黄色为阳性，注意排除胞质非特异性着色（可结合Pax-8联合检测甲状腺来源）。

WB注意事项：

样本处理：肺/甲状腺肿瘤组织需用含蛋白酶抑制剂的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

抗体稀释：一抗推荐1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP标记二抗（抗小鼠IgG）1:5000室温孵育1小时。
