产品介绍Gamma-aminobutyric acid A receptors [GABA(A) receptors] are ligand-gated chloride channels that mediate inhibitory neurotransmission. This gene encodes GABA(A) receptor-associated protein, which is highly positively charged in its N-terminus and shares sequence similarity with light chain-3 of microtubule-associated proteins 1A and 1B. This protein clusters neurotransmitter receptors by mediating interaction with the cytoskeleton. [provided by RefSeq, Jul 2008]

靶点与功能

Galectin-3（Gal-3）是半乳糖凝集素家族**含胶原蛋白样结构域的成员，分子量约30kDa，通过C端碳水化合物识别域（CRD）结合β-半乳糖苷，N端重复序列介导多聚体形成。其核心功能包括：

病理作用：促进肿瘤细胞侵袭转移（上调MMP-9）、心肌纤维化（激活TGF-β通路）、炎症反应（募集巨噬细胞）；

临床价值：血清Gal-3是心力衰竭预后独立标志物（正常参考值<17.8ng/mL），也是甲状腺滤泡癌与腺瘤鉴别的关键指标（癌组织中核定位阳性）；

研究意义：通过抗体阻断Gal-3可抑制肿瘤血管生成，是纤维化和癌症治疗的潜在靶点。

实验操作关键要点

IHC-P组织染色（甲状腺/心脏样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），甲状腺癌样本需检测核/质定位（核阳性提示恶性）；

阳性对照：选择乳腺癌组织（高表达）或肝纤维化组织（胞质强阳性）；

显色控制：DAB显色5-8分钟，避免过染导致背景干扰。

WB验证（细胞裂解液）：

样本处理：使用含1mM PMSF的RIPA裂解液（避免蛋白酶降解），12000g离心15分钟取上清；

电泳条件：12% SDS-PAGE凝胶，恒压120V分离胶，转膜用0.45μm PVDF膜（250mA恒流1小时）；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗1:5000室温孵育1小时。

ELISA定量检测（血清样本）：

包被条件：抗体用碳酸盐缓冲液（pH9.6）稀释至2μg/mL，4℃包被16小时；

样本处理：血清1:10稀释（含1% BSA的PBS），37℃孵育2小时；

标准曲线：0.1-100ng/mL重组Gal-3蛋白，R²需≥0.99。
