产品介绍 is one synthetic peptide derived from human Galectin 3.

Galactose-specific lectin which binds IgE. May mediate with the alpha-3, beta-1 integrin the stimulation by CSPG4 of endothelial cells migration. Together with DMBT1, required for terminal differentiation of columnar epithelial cells during early embryogenesis. In the nucleus: acts as a pre-mRNA splicing factor. Involved in acute inflammatory responses including neutrophil activation and adhesion, chemoattraction of monocytes macrophages, opsonization of apoptotic neutrophils, and activation of mast cells.

靶点与功能

半乳糖凝集素3（Galectin-3，Gal-3）是β-半乳糖苷结合蛋白，分子量约29kDa，通过以下机制参与病理生理过程：

炎症与纤维化：通过识别细胞表面糖链（如N-聚糖）激活TLR4/NF-κB通路，促进促炎因子（TNF-α、IL-6）释放，在肝纤维化、肺纤维化中作为关键驱动因子；

肿瘤进展：通过与整合素β1结合增强肿瘤细胞侵袭能力，同时抑制T细胞抗肿瘤免疫，与乳腺癌、结直肠癌转移风险正相关；

心脏疾病：心肌梗死模型中Gal-3可促进心肌成纤维细胞活化，是心力衰竭预后评估的独立标志物（血清水平＞17ng/mL提示高风险）。

实验操作关键要点

IHC-P肝纤维化组织染色：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），冷却后用3% H₂O₂阻断内源性过氧化物酶；

阳性定位：肝星状细胞胞质/胞核阳性（激活态星状细胞核定位增强），汇管区炎症细胞也可检测到胞质信号；

染色优化：一抗推荐1:400稀释（5% BSA+TBST），4℃孵育过夜，DAB显色3分钟，苏木素复染细胞核（区分星状细胞与肝细胞）。

WB检测（巨噬细胞样本）：

样本处理：LPS刺激RAW264.7细胞24小时（诱导Gal-3表达），使用含1mM PMSF的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜0.45μm PVDF膜（250mA恒流1.5小时）；

抗体孵育：抗体1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗1:5000室温孵育1小时，曝光时间20秒（29kDa单体条带清晰）。

IF双标实验（肿瘤微环境）：

推荐与CD206抗体共染（M2型巨噬细胞标志物），使用Alexa Fluor 488（绿色，Gal-3）和594（红色，CD206），共聚焦观察二者在肿瘤间质的共定位（提示免疫抑制微环境）。
