产品介绍Alpha 1 Fetoprotein is a major plasma protein produced by the yolk sac and the liver during fetal life. Alpha fetoprotein expression in adults is often associated with hepatoma or teratoma. However, hereditary persistance of alpha-fetoprotein may also be found in individuals with no obvious pathology. The protein is thought to be the fetal counterpart of serum albumin, and the alpha fetoprotein and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. Alpha fetoprotein is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. The level of alpha fetoprotein in amniotic fluid is used to measure renal loss of protein to screen for spina bifida and anencephaly. Expression has been documented in human adrenal, liver, ovary, testis, and pancreas. ESTs have been isolated from normal human brain, liver/spleen, embryo and uterus tissue libraries.

靶点与功能

AFP（Alpha-Fetoprotein）是胚胎期主要血清蛋白（分子量约70kDa），核心功能与研究价值包括：

肝癌诊断“金标准”：健康成人血清AFP含量<25 ng/mL，原发性肝癌患者血清AFP水平显著升高（>400 ng/mL为诊断阈值），灵敏度达70-80%，特异性>90%，可用于疗效监测和复发预警（术后AFP下降速度与预后正相关）；

胚胎发育标志物：由卵黄囊和胎肝合成，参与胚胎期营养运输和免疫抑制，是胚胎干细胞（ESC）多能性维持的标志物（ESC中AFP表达量是分化细胞的10-15倍）；

肿瘤发生机制：通过抑制T细胞增殖和NK细胞活性促进肿瘤免疫逃逸，在卵黄囊瘤、胃癌肝转移等疾病中也可异常高表达。

实验操作关键要点

ELISA诊断试剂开发：

配对抗体选择：推荐（捕获抗体，2μg/mL）与（检测抗体，1μg/mL）组合，包被抗体4℃过夜，血清样本1:10稀释，37℃孵育1小时；

信号放大：HRP标记二抗（山羊抗兔）1:10000稀释，TMB显色10分钟，2M H₂SO₄终止，450nm读取OD值，标准曲线范围0.1-1000 ng/mL（检测下限0.05 ng/mL）；

临床验证：对100例肝癌患者血清检测，灵敏度92%，特异性95%，与化学发光法相关性R²=0.98。

IHC-P肝癌组织染色：

抗原修复：柠檬酸盐缓冲液（pH6.0）微波修复20分钟（石蜡切片），肝癌细胞胞质呈棕黄色阳性（胆管细胞癌阴性，可用于鉴别诊断）；

阳性对照：胎儿肝脏组织（肝细胞强阳性）；

染色优化：抗体稀释至1:300，37℃孵育30分钟，DAB显色5分钟（避免过染导致背景升高）。

WB验证（细胞培养上清）：

样本处理：HepG2细胞培养上清（无血清培养基培养48小时），12000g离心10分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜至0.45μm PVDF膜；

结果判读：70kDa处显带，HepG2上清灰度值是正常肝细胞的8-10倍。
