产品介绍Alpha 1 Fetoprotein is a major plasma protein produced by the yolk sac and the liver during fetal life. Alpha fetoprotein expression in adults is often associated with hepatoma or teratoma. However, hereditary persistance of alpha-fetoprotein may also be found in individuals with no obvious pathology. The protein is thought to be the fetal counterpart of serum albumin, and the alpha fetoprotein and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. Alpha fetoprotein is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. The level of alpha fetoprotein in amniotic fluid is used to measure renal loss of protein to screen for spina bifida and anencephaly. Expression has been documented in human adrenal, liver, ovary, testis, and pancreas. ESTs have been isolated from normal human brain, liver/spleen, embryo and uterus tissue libraries.

AFP（甲胎蛋白）是胚胎期肝脏和卵黄囊合成的糖蛋白（分子量69kDa），核心应用价值包括：

临床诊断：成人血清AFP正常参考值＜20ng/mL，原发性肝癌患者阳性率＞70%（浓度可达200-1000ng/mL），是肝癌筛查、疗效监测及复发预警的**指标；

包被实验操作关键要点

ELISA板包被优化：

缓冲液选择：0.05M碳酸盐缓冲液（pH9.6）溶解抗体，包被浓度推荐 2μg/mL（每孔100μL），4℃静置16小时；

封闭条件：5%脱脂奶粉（PBS-T配制）37℃封闭2小时，有效减少非特异性结合；

洗涤步骤：PBST（含0.05% Tween-20）洗涤3次，每次3分钟，确保去除未结合抗体。

胶体金试纸条包被：

NC膜活化：1% BSA-PBS预处理30分钟，晾干后使用；

划线参数：包被抗体浓度1mg/mL，划膜速度30mm/s，线宽0.8mm，37℃烘干2小时；

配对抗体：推荐与检测抗体（如克隆号A4）组合，形成“包被抗体捕获AFP-检测抗体标记胶体金”双抗夹心体系。

稳定性验证：

37℃加速实验7天，包被板ELISA信号衰减＜10%；

反复冻融3次后，抗体活性保留率＞90%（建议分装为50μL/管，避免反复冻融）。
