Actin filaments can form both stable and labile structures and are crucial components of microvilli and the contractile apparatus of muscle cells. While lower eukaryotes, such as yeast, have only one Actin gene, higher eukaryotes have several isoforms encoded by a family of genes. At least six types of Actin are present in mammalian tissues and fall into three classes. alpha-Actin expression is limited to various types of muscle, whereas beta- and gamma-Actin are the principle constituents of filaments in other tissues. Members of the small GTPase family regulate the organization of the Actin cytoskeleton. Rho controls the assembly of Actin stress fibers and focal adhesion. Rac regulates Actin filament accumulation at the plasma membrane. Cdc42 stimulates formation of filopodia.

PubMedPubMed 靶点与功能 α-SMA(平滑肌肌动蛋白α2,ACTA2基因编码)是肌动蛋白家族的收缩型异构体(分子量42kDa),核心功能与研究价值包括: 细胞表型标志物:特异性表达于血管平滑肌细胞、胃肠道平滑肌细胞及活化肌成纤维细胞,静息成纤维细胞阴性,是区分肌成纤维细胞与普通成纤维细胞的**指标; 纤维化驱动因子:通过与肌球蛋白结合形成应力纤维,增强细胞收缩能力,促进细胞外基质(ECM)沉积(如肝纤维化中α-SMA⁺细胞数量与胶原含量正相关); 肿瘤微环境调控:在乳腺癌、胰腺癌等肿瘤间质中高表达,通过“基质硬化”促进肿瘤侵袭转移(α-SMA⁺肌成纤维细胞密度是胰腺癌预后不良的独立危险因素)。 实验操作关键要点 IHC-P纤维化组织染色: 抗原修复:柠檬酸盐缓冲液(pH6.0)微波修复20分钟(石蜡切片),肝纤维化组织中汇管区肌成纤维细胞呈棕黄色阳性(正常肝组织仅血管壁阳性); 阳性对照:大鼠子宫平滑肌组织(胞质弥漫强阳性); 定量分析:采用Image-Pro Plus计算α-SMA⁺面积占比(肝纤维化S2期占比>15%,S4期可达40%以上)。 WB肌成纤维细胞活化检测: 样本处理:TGF-β1(10ng/mL)刺激L929细胞48小时,RIPA裂解液(含1mM PMSF)冰上裂解30分钟; 电泳条件:12% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,转膜使用0.45μm PVDF膜; 抗体稀释:抗体1:2000稀释(5%脱脂奶粉+TBST),4℃孵育过夜,HRP二抗1:5000室温孵育1小时,曝光时间30秒。 IF细胞骨架共定位: 与波形蛋白(Vimentin) 共染,α-SMA(红色)在肌成纤维细胞中形成“束状应力纤维”,与Vimentin(绿色)部分共定位(重叠系数>0.6提示细胞活化)。 公司正在出售的产品
