PubMedPubMed 产品介绍The protein encoded by this gene is a member of the keratin family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21. This gene encodes the type I intermediate filament chain keratin 17, expressed in nail bed, hair follicle, sebaceous glands, and other epidermal appendages. Mutations in this gene lead to Jackson-Lawler type pachyonychia congenita and steatocystoma multiplex. [provided by RefSeq, Aug 2008]. 靶点与功能

IHC-P病理诊断(石蜡切片): 抗原修复:采用柠檬酸钠缓冲液(pH6.0) 微波修复20分钟(95℃),冷却后用3% H₂O₂封闭内源性过氧化物酶10分钟; 阳性定位:肿瘤细胞胞质呈棕黄色细丝状阳性,鳞癌组织中呈弥漫分布,腺癌组织中阴性; 阳性对照:推荐使用食管鳞癌组织或皮肤鳞癌组织,确保抗体活性。 WB验证(细胞/组织裂解液): 样本处理:使用含1mM PMSF的RIPA裂解液冰上裂解30分钟,12000g离心15分钟取上清; 电泳条件:10% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,转膜选用0.45μm PVDF膜(250mA恒流1小时); 抗体孵育:一抗1:500稀释(5%脱脂奶粉+TBST),4℃孵育过夜,HRP二抗(山羊抗小鼠)1:5000室温孵育1小时。 公司正在出售的产品
