PubMedPubMed 产品介绍Cytokeratins are a subfamily of intermediate filament proteins and are characterized by a remarkable biochemical diversity, represented in human epithelial tissues by at least 20 different polypeptides. They range in molecular weight between 40 kDa and 68 kDa and isoelectric pH between 4.9 – 7.8. The individual human cytokeratins are designated 1 to 20. The various epithelia in the human body usually express cytokeratins which are not only characteristic of the type of epithelium, but also related to the degree of maturation or differentiation within an epithelium. Cytokeratin subtype expression patterns are used to an increasing extent in the distinction of different types of epithelial malignancies. The cytokeratin antibodies are not only of assistance in the differential diagnosis of tumors using immunohistochemistry on tissue sections, but are also a useful tool in cytopathology and flow cytometric assays. 靶点与功能
细胞角蛋白7(Cytokeratin 7, CK7)是II型中性角蛋白,分子量约54kDa,主要表达于腺上皮细胞(如肺、乳腺、卵巢、胆管上皮)。其核心功能与临床价值包括: 组织特异性:正常情况下在肺、乳腺、胆管等上皮细胞表达,与CK20联合检测可用于肿瘤原发灶判断(如CK7⁺/CK20⁻提示肺/乳腺来源); 肿瘤诊断:肺腺癌(阳性率90%)、卵巢癌(100%)、胆管癌(95%)中高表达,是腺癌与鳞癌鉴别的重要标志物; 研究意义:在间皮瘤与腺癌的鉴别中具有特异性(间皮瘤CK7⁺但不表达BerEP4)。 实验操作关键要点
IHC-P肿瘤分型(肺腺癌样本): 抗原修复:柠檬酸钠缓冲液(pH6.0)微波修复20分钟(95℃),冷却后用3% H₂O₂封闭10分钟; 阳性定位:肿瘤细胞胞质呈弥漫性棕黄色阳性,鳞癌组织阴性; 对照设置:必须包含肺腺癌(阳性) 和肺鳞癌(阴性) 组织对照,确保抗体特异性。 WB验证(细胞裂解液): 样本处理:使用含1mM PMSF的RIPA裂解液冰上裂解30分钟,12000g离心15分钟取上清; 电泳条件:10% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,转膜选用0.45μm PVDF膜(250mA恒流1.5小时); 抗体孵育:一抗1:1000稀释(5%脱脂奶粉+TBST),4℃孵育过夜,HRP二抗(山羊抗小鼠)1:5000室温孵育1小时。 公司正在出售的产品
