PubMedPubMed 产品介绍 The collagens are a superfamily of proteins that play a role in maintaining the integrity of various tissues. Collagens are extracellular matrix proteins and have a triple-helical domain as their common structural element. Collagen VI is a major structural component of microfibrils. The basic structural unit of collagen VI is a heterotrimer of the alpha1(VI), alpha2(VI), and alpha3(VI) chains. The alpha2(VI) and alpha3(VI) chains are encoded by the COL6A2 and COL6A3 genes, respectively. The protein encoded by this gene is the alpha 1 subunit of type VI collagen (alpha1(VI) chain). Mutations in the genes that code for the collagen VI subunits result in the autosomal dominant disorder, Bethlem myopathy. [provided by RefSeq, Jul 2008] 靶点与功能

WB优化(纤维化组织样本): 样本处理:使用含蛋白酶抑制剂(1mM PMSF) 的RIPA裂解液冰上裂解30分钟,12000g离心取上清;若检测多聚体,需避免剧烈涡旋; 电泳条件:8% SDS-PAGE凝胶(因分子量较大),恒压80V浓缩胶、120V分离胶,目标条带约106kDa(单体)或更高(多聚体); 抗体孵育:一抗1:2000稀释(5% BSA+TBST),4℃摇床孵育过夜,HRP二抗(山羊抗兔)1:5000室温孵育1小时。 IHC-P注意事项(皮肤组织切片): 抗原修复:采用柠檬酸钠缓冲液(pH6.0) 微波修复15分钟(95℃),增强细胞外基质信号; 阳性定位:细胞外基质呈网状或条索状棕黄色阳性,皮肤真皮层、肌肉筋膜区信号最强; 封闭处理:5%驴血清+0.3% Triton X-100室温封闭45分钟,减少非特异性结合。 公司正在出售的产品
