MECT1 does not appear to modulate CREB1 DNA-binding activity but enhances the interaction of CREB1 with TAF4/TAFII-130. MECT1 translocates with MAML2 (MasterMind-Like Protein 2) to yield a fusion oncogene: t(11;19) (q21;p13). This translocation occurs in mucoepidermoid carcinomas, benign Warthin tumors and clear cell hidradenomas. The novel fusion product that results disrupts the Notch signaling pathway. The fusion protein consists of the N-terminus of MECT1 joined to the C-terminus of MAML2. The reciprocal fusion protein consisting of the N-terminus of MAML2 joined to the C-terminus of MECT1 has been detected in a small number of mucoepidermoid carcinomas. WB优化(神经组织样本): 样本处理:使用含蛋白酶抑制剂(1mM PMSF) 的RIPA裂解液冰上裂解30分钟,12000g离心取上清;若检测磷酸化修饰,需添加磷酸酶抑制剂; 电泳条件:8% SDS-PAGE凝胶(因分子量较大),恒压80V浓缩胶、120V分离胶,目标条带约106kDa; 抗体孵育:单克隆抗体推荐1:1500稀释(5% BSA+TBST),4℃孵育过夜,HRP二抗(山羊抗兔)1:5000室温孵育1小时。 IHC-P注意事项(脑组织切片): 抗原修复:采用柠檬酸钠缓冲液(pH6.0) 微波修复15分钟(95℃),自然冷却至室温; 阳性定位:细胞膜/胞质呈棕黄色颗粒状阳性,在胚胎神经管、肾小球足细胞中高表达; 封闭处理:5%正常山羊血清+0.3% Triton X-100室温封闭45分钟,降低非特异性结合。 公司正在出售的产品
