PubMedPubMed 产品介绍Cytochrome c oxidase (COX) is the terminal enzyme of the mitochondrial respiratory chain. It is a multi-subunit enzyme complex that couples the transfer of electrons from cytochrome c to molecular oxygen and contributes to a proton electrochemical gradient across the inner mitochondrial membrane. The complex consists of 13 mitochondrial- and nuclear-encoded subunits. The mitochondrially-encoded subunits perform the electron transfer and proton pumping activities. The functions of the nuclear-encoded subunits are unknown but they may play a role in the regulation and assembly of the complex. This gene encodes the nuclear-encoded subunit IV isoform 1 of the human mitochondrial respiratory chain enzyme. It is located at the 3' of the NOC4 (neighbor of COX4) gene in a head-to-head orientation, and shares a promoter with it. [provided by RefSeq, Jul 2008] 靶点与功能

WB内参校正(线粒体蛋白样本): 样本处理:线粒体提取需使用差速离心法(线粒体提取试剂盒),避免胞质蛋白污染; 电泳条件:15% SDS-PAGE凝胶(分离小分子量蛋白),恒压80V浓缩胶、120V分离胶,转膜用0.22μm PVDF膜(150mA恒流1小时); 抗体稀释:一抗1:2000稀释(5%脱脂奶粉+TBST),4℃孵育过夜,HRP二抗1:5000室温孵育1小时,曝光时间5-10秒(避免过曝)。 IHC线粒体定位(心肌组织样本): 抗原修复:柠檬酸钠缓冲液(pH6.0)微波修复10分钟(95℃),冷却后用3% H₂O₂封闭10分钟; 阳性定位:心肌细胞胞质内呈颗粒状阳性(线粒体分布区域),与细胞核无共定位; 对照设置:必须包含COX IV敲低细胞系(阴性对照)和正常组织(阳性对照)。 公司正在出售的产品
