商品属性 产品介绍The protein encoded by this gene belongs to the aldehyde dehydrogenase family. Aldehyde dehydrogenase is the next enzyme after alcohol dehydrogenase in the major pathway of alcohol metabolism. There are two major aldehyde dehydrogenase isozymes in the liver, cytosolic and mitochondrial, which are encoded by distinct genes, and can be distinguished by their electrophoretic mobility, kinetic properties, and subcellular localization. This gene encodes the cytosolic isozyme. Studies in mice show that through its role in retinol metabolism, this gene may also be involved in the regulation of the metabolic responses to high-fat diet. [provided by RefSeq, Mar 2011].

PubMedPubMed 靶点与功能 ALDH1(Aldehyde Dehydrogenase 1 Family Member A1)是NAD⁺依赖的醛脱氢酶超家族成员(分子量约55kDa),核心功能与研究价值包括: 代谢通路枢纽:催化乙醛氧化为乙酸(Km=0.15mM),是酒精代谢的关键限速酶,ALDH1活性缺失(如东亚人群常见的ALDH2*2突变)会导致乙醛蓄积,增加酒精性肝损伤风险; 肿瘤干细胞标志物:在乳腺癌、结直肠癌、肝癌等肿瘤中,ALDH1⁺细胞亚群具有高致瘤性(成球能力是ALDH1⁻细胞的8-10倍),与化疗耐药和复发正相关; 视黄酸合成:通过将视黄醛氧化为视黄酸,调控胚胎发育和干细胞分化(如神经发生、造血干细胞维持)。 实验操作关键要点 WB肿瘤干细胞检测: 样本处理:乳腺癌MCF-7细胞用含1mM DTT的RIPA裂解液冰上裂解30分钟,12000g离心15分钟取上清;ALDH1⁺亚群可通过ALDEFLUOR试剂盒分选(阳性率约1-5%); 电泳条件:10% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,电泳90分钟; 抗体孵育:重组单抗1:1500稀释(5% BSA+TBST),4℃孵育过夜,HRP二抗(山羊抗兔)1:5000室温孵育1小时,曝光时间30-60秒。 IHC-P肿瘤组织染色: 抗原修复:柠檬酸盐缓冲液(pH6.0)微波修复20分钟(石蜡切片),肿瘤细胞胞质呈棕黄色阳性(癌巢边缘细胞染色更强); 阳性对照:正常肝脏组织(肝细胞胞质弱阳性); 联合检测:与CD44抗体共染,ALDH1⁺/CD44⁺双阳性细胞比例>10%提示高转移风险。 IF干细胞分化监测: 诱导多能干细胞(iPSC)向神经细胞分化7天后,ALDH1(绿色)与 Nestin(红色)共定位,荧光强度随分化进程逐渐增强(第14天达峰值)。 公司正在出售的产品
