靶点与功能

PSA（Prostate-Specific Antigen）是由前列腺上皮细胞分泌的丝氨酸蛋白酶，分子量约26kDa，含261个氨基酸（1-24位为信号肽，第69位天冬酰胺为糖基化位点）。其核心功能包括：

生理作用：参与精液液化（降解精囊腺分泌的凝固蛋白）；

临床价值：血清PSA水平是前列腺癌筛查的**标志物，正常参考值≤4ng/mL，4-10ng/mL为“灰区”，需结合fPSA/tPSA比值（比值＜0.15提示恶性风险）；

研究意义：探索PSA在肿瘤微环境中的作用（如促进血管生成、抑制抗肿瘤免疫），是开发新型诊疗技术的基础。

实验操作关键要点

ELISA定量检测（血清样本）：

包被优化：抗体用碳酸盐缓冲液（pH9.6）稀释至2μg/mL，4℃包被过夜，PBST洗涤3次（每次5分钟）；

样本处理：血清需56℃灭活30分钟（去除补体干扰），用含1% BSA的PBS按1:100稀释，37℃孵育1.5小时；

信号放大：HRP标记二抗（山羊抗兔）1:5000稀释，TMB显色15分钟，2M H₂SO₄终止，450nm读取OD值（标准曲线范围0.1-100ng/mL）。

IHC-P组织定位（前列腺癌样本）：

抗原修复：采用EDTA缓冲液（pH9.0） 高压修复10分钟（石蜡切片），抗原表位暴露更充分；

阳性对照：正常前列腺组织腺上皮细胞胞质呈棕黄色阳性，癌组织中表达增强且分布不均；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭1小时，降低非特异性结合（PSA易与基底细胞产生交叉反应）。

WB验证（细胞裂解液）：

样本制备：LNCaP细胞（高表达PSA）用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜采用0.45μm PVDF膜（300mA恒流1.5小时）；

抗体孵育：一抗1:1000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，曝光时间30-60秒（避免信号过强）。
