产品介绍The nuclear lamina consists of a two-dimensional matrix of proteins located next to the inner nuclear membrane. The lamin family of proteins make up the matrix and are highly conserved in evolution. During mitosis, the lamina matrix is reversibly disassembled as the lamin proteins are phosphorylated. Lamin proteins are thought to be involved in nuclear stability, chromatin structure and gene expression. Vertebrate lamins consist of two types, A and B. This gene encodes one of the two B type proteins, B1. Alternative splicing results in transcript variants and a duplication of this gene is associated with autosomal dominant adult-onset leukodystrophy (ADLD). [provided by RefSeq, Oct 2010].

靶点与功能

核纤层蛋白B（Lamin B）是中间丝蛋白家族成员，主要包括Lamin B1（68kDa）和Lamin B2（66kDa）两种亚型，通过C端异戊二烯化锚定在内核膜，形成核纤层网络。其核心功能包括：

核结构维持：为细胞核提供机械支撑，参与核膜组装与染色质组织；

细胞周期调控：在有丝分裂期核膜破裂与重建中起关键作用；

疾病关联：Lamin B1缺失与早老症（Hutchinson-Gilford综合征）、神经退行性疾病及肿瘤侵袭转移密切相关，是核结构异常的重要标志物。

实验操作关键要点

IF核膜定位实验（贴壁细胞）：

细胞处理：HeLa细胞用4%多聚甲醛固定15分钟，0.2% Triton X-100透化20分钟，PBS洗涤3次；

染色步骤：一抗1:200稀释（含1% BSA的PBS）4℃孵育过夜，Alexa Fluor 488标记二抗（绿色）室温孵育1小时，DAPI（蓝色）染核，激光共聚焦观察细胞核膜环状阳性信号（与DAPI边缘重叠）。

WB检测（细胞裂解液）：

样本制备：293T细胞用含1mM PMSF的RIPA裂解液冰上裂解30分钟，12000g离心取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜使用0.45μm PVDF膜（恒流300mA 90分钟）；

抗体孵育：一抗1:1000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗1:5000室温孵育1小时，曝光30-60秒（目标条带：Lamin B1约68kDa，Lamin B2约66kDa）。
