PubMedPubMed 产品介绍The protein encoded by this gene is a bifunctional enzyme that catalyzes the oxidative conversion of delta(5)-ene-3-beta-hydroxy steroid, and the oxidative conversion of ketosteroids. It plays a crucial role in the biosynthesis of all classes of hormonal steroids. This gene is predominantly expressed in the adrenals and the gonads. Mutations in this gene are associated with 3-beta-hydroxysteroid dehydrogenase, type II, deficiency. Alternatively spliced transcript variants have been found for this gene. IHC-P组织染色(胎盘或肿瘤样本): 抗原修复:采用柠檬酸钠缓冲液(pH6.0) 微波修复20分钟(95℃),石蜡切片需脱蜡至水后进行; 阳性定位:滋养层细胞胞质呈棕黄色颗粒状阳性,正常胎盘绒毛滋养层细胞高表达,葡萄胎组织中表达增强; 对照设置:需包含正常胎盘组织(阳性对照)和膀胱组织(阴性对照,排除非特异性结合)。 WB验证(细胞裂解液): 样本处理:滋养层细胞株(如JEG-3)用含蛋白酶抑制剂(1mM PMSF) 的RIPA裂解液冰上裂解30分钟,12000g离心15分钟取上清; 电泳条件:10% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,转膜使用0.45μm PVDF膜(250mA恒流1.5小时); 抗体孵育:一抗1:1000稀释(5%脱脂奶粉+TBST),4℃孵育过夜,HRP标记二抗(山羊抗兔)1:5000室温孵育1小时,曝光时间30-60秒。 公司正在出售的产品
