产品介绍This gene encodes a cell surface glycoprotein and member of the immunoglobulin superfamily of proteins. The encoded protein is involved in cell adhesion and cell communication in numerous cell types, but particularly in cells of the immune and nervous systems. The encoded protein is widely used as a marker for hematopoietic stem cells. This gene may function as a tumor suppressor in nasopharyngeal carcinoma. 靶点与功能

CD90（又称Thy1）是一种25-35kDa的GPI锚定糖蛋白，属于免疫球蛋白超家族，核心功能包括：

干细胞标志物：特异性表达于骨髓间充质干细胞（MSCs）、神经干细胞表面，是MSCs分离纯化的经典标志（如CD90⁺CD44⁺CD34⁻表型）；

神经发育调控：参与轴突生长和突触形成，在成熟神经元轴突末端高表达；

肿瘤微环境作用：在前列腺癌、肝癌等肿瘤基质细胞中高表达，与肿瘤侵袭转移相关。

实验操作关键要点

MSCs流式鉴定（FC）：

抗体组合：推荐与CD44、CD34抗体联合标记（CD90⁺CD44⁺CD34⁻为MSCs特征表型），荧光标记抗体（如PerCP/Cyanine5.5偶联）需提前优化浓度（2μl/test）；

样本处理：胰酶消化的MSCs用含2% FBS的PBS洗涤2次，4℃避光孵育30分钟，避免固定剂影响GPI锚定蛋白活性。

WB检测（神经组织样本）：

样本制备：大鼠脑组织用含1% NP-40的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清，推荐上样量30-50μg总蛋白；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜选用0.45μm PVDF膜（250mA恒流1.5小时）。
