产品介绍IgM normally constitutes about 10% of serum immunoglobulins. IgM antibody is prominent in early immune responses to most antigens and is largely confined to plasma due to it's large size. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. Due to it's high valency IgM is more efficient than other isotypes is binding antigens with repeating epitopes (virus particles and red blood cells) and is more efficient than IgG in activiating the complement pathway. The gene for the mu constant region contains four domains separated by short intervening sequences.

人免疫球蛋白M（IgM）是分子量约970kDa的五聚体蛋白，由5个单体通过J链连接，主要存在于血清（占总Ig的5%-10%）和B细胞表面（单体形式）。其核心功能包括：

免疫防御：作为初次免疫应答的主要抗体，通过高亲和力结合病原体（如病毒、细菌）激活补体经典途径，启动吞噬作用；

临床价值：血清IgM水平升高提示急性感染（如EB病毒、甲型肝炎）或自身免疫病，是疾病早期诊断的关键标志物；

研究意义：重组兔单抗可特异性识别IgM重链（μ链），避免与其他免疫球蛋白（IgG、IgA）交叉反应，适用于精准定量检测与免疫沉淀实验。

实验操作关键要点

ELISA定量检测（血清/血浆样本）：

包被优化：抗体用0.05M碳酸盐缓冲液（pH9.6）按2μg/mL稀释，4℃包被过夜，PBST洗涤3次；

样本处理：血清需1:100稀释（含1% BSA的PBS），37℃孵育1小时，避免溶血样本干扰（血红蛋白会非特异性结合抗体）；

显色控制：HRP标记二抗（山羊抗兔IgG）1:5000稀释，TMB显色10分钟，2M H₂SO₄终止反应，450nm读取OD值（标准曲线范围0.1-200ng/mL）。

WB验证（IgM标准品或细胞裂解液）：

样本制备：纯IgM标准品直接上样（0.5-2μg/孔），B细胞裂解液需用含蛋白酶抑制剂的RIPA冰上裂解30分钟；

电泳条件：6% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、100V分离胶，转膜使用0.45μm PVDF膜（恒流300mA 90分钟）；

抗体孵育：一抗1:1000稀释（5%脱脂奶粉+TBST），4℃摇床孵育过夜，二抗1:5000室温孵育1小时，曝光1-2分钟（目标条带约970kDa五聚体及160kDa单体）。
