PubMedPubMed 产品介绍This gene encodes a member of the heat shock protein 70 family, which contains both heat-inducible and constitutively expressed members. This protein belongs to the latter group, which are also referred to as heat-shock cognate proteins. It functions as a chaperone, and binds to nascent polypeptides to facilitate correct folding. It also functions as an ATPase in the disassembly of clathrin-coated vesicles during transport of membrane components through the cell. WB检测(细胞裂解液): 样本处理:使用含1mM ATP的RIPA裂解液(维持HSP71活性)冰上裂解30分钟,12000g离心15分钟取上清; 电泳条件:10% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,转膜用0.45μm PVDF膜(250mA恒流1.5小时); 抗体孵育:小鼠单抗1:8000稀释(5%脱脂奶粉+TBST),4℃孵育过夜,HRP二抗(山羊抗小鼠)1:5000室温孵育1小时。 IHC-P组织染色(肿瘤样本): 抗原修复:采用柠檬酸钠缓冲液(pH6.0) 微波修复20分钟(95℃),冷却后用PBS洗3次; 阳性对照:推荐使用热应激处理的HepG2细胞爬片(胞质强阳性); 显色控制:DAB显色5-8分钟,阳性信号为棕黄色颗粒(肿瘤细胞胞质弥漫性表达)。 公司正在出售的产品
