产品介绍The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains an extracellular domain, a single transmembrane segment and two tandem intracytoplasmic catalytic domains, and thus belongs to receptor type PTP. This gene is specifically expressed in hematopoietic cells. This PTP has been shown to be an essential regulator of T- and B-cell antigen receptor signaling. It functions through either direct interaction with components of the antigen receptor complexes, or by activating various Src family kinases required for the antigen receptor signaling. This PTP also suppresses JAK kinases, and thus functions as a regulator of cytokine receptor signaling. Four alternatively spliced transcripts variants of this gene, which encode distinct isoforms, have been reported. [provided by RefSeq, Jul 2008].

靶点与功能

CD45（Cluster of Differentiation 45）是跨膜蛋白酪氨酸磷酸酶，表达于所有造血细胞（除成熟红细胞和血小板），通过去磷酸化Lck/Fyn等Src家族激酶调控T/B细胞活化信号。其不同剪接异构体（CD45RA/RO/RC）是区分初始T细胞（CD45RA⁺）与记忆T细胞（CD45RO⁺） 的关键标志物，在自身免疫病（如多发性硬化）、肿瘤免疫治疗（CAR-T细胞表型分析）中应用广泛。

实验操作关键要点

流式细胞术（外周血T细胞分型）：

样本处理：EDTA抗凝血经红细胞裂解液（BD Pharm Lyse） 处理5分钟，PBS洗涤2次，调整细胞浓度至1×10⁶/mL；

染色条件：CD45-FITC（BD HI30）与CD3-PE（1:50）避光孵育30分钟（4℃），2%多聚甲醛固定后上样；

数据解读：CD45⁺CD3⁺设门为T细胞群，进一步分析CD45RA/RO表达区分初始/记忆亚群（建议使用FlowJo软件）。

IHC-P组织染色（淋巴瘤样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 高温高压修复2分钟（121℃），自然冷却；

阳性定位：淋巴细胞胞膜呈棕黄色线性阳性，肿瘤组织中CD45高表达（与上皮源性肿瘤阴性对照区分）；

对照设置：必须包含正常淋巴结组织（阳性对照）和胃癌组织（阴性对照，上皮细胞不表达CD45）。

WB验证（Jurkat细胞裂解液）：

样本制备：使用含1mM Na₃VO₄（磷酸酶抑制剂） 的RIPA裂解液，冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：8% SDS-PAGE凝胶（低浓度胶分离高分子量蛋白），恒压80V浓缩胶、100V分离胶，转膜采用0.45μm PVDF膜（300mA恒流90分钟）；

抗体孵育：兔单抗1:2000稀释（5% BSA+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间30-60秒。
