产品介绍This gene encodes a member of the scavenger receptor cysteine-rich (SRCR) superfamily. Members of this family are secreted or membrane-anchored proteins mainly found in cells associated with the immune system. This protein is a type-I transmembrane glycoprotein found on the surface of thymocytes, T lymphocytes and a subset of B lymphocytes. The encoded protein contains three SRCR domains and may act as a receptor to regulate T-cell proliferation. Alternative splicing results in multiple transcript variants encoding different isoforms. 靶点与功能

CD5是跨膜糖蛋白，主要表达于95%胸腺细胞、成熟T细胞及B-1a B细胞亚群（约5%外周B细胞），通过调控TCR/BCR信号通路阈值参与免疫耐受和自身免疫反应。其异常表达与T细胞淋巴瘤（如套细胞淋巴瘤CD5⁺）、慢性淋巴细胞白血病（CLL） 密切相关，是淋巴瘤病理诊断的重要标志物（例如区分套细胞淋巴瘤与滤泡中心淋巴瘤，后者CD5⁻）。

实验操作关键要点

WB实验（Jurkat T细胞裂解液）：

样本处理：使用含1mM PMSF的RIPA裂解液冰上裂解20分钟，12000g离心15分钟取上清，上样量推荐30-50μg；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜采用0.45μm PVDF膜（250mA恒流60分钟）；

抗体孵育：一抗1:2000稀释（5%脱脂奶粉+TBST），4℃孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间15-30秒（预期条带67kDa）。

流式细胞术（外周血T细胞检测）：

样本制备：EDTA抗凝血经红细胞裂解液处理后，PBS洗涤2次，调整细胞浓度至1×10⁶/mL；

染色步骤：FITC标记CD5抗体（1:50稀释）避光孵育20分钟（4℃），无需洗涤直接上样（避免荧光淬灭）；

设门策略：先通过FSC/SSC设门淋巴细胞群，再以CD5⁺CD3⁺双阳性定义T细胞亚群（正常外周血T细胞CD5阳性率>90%）。

IHC-P（淋巴瘤组织切片）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复15分钟（95℃），自然冷却；

阳性定位：T细胞胞膜呈棕黄色线性着色，套细胞淋巴瘤可见肿瘤细胞弥漫性膜阳性，而滤泡性淋巴瘤阴性；

对照设置：以正常扁桃体组织为阳性对照（T细胞区棕黄色膜染色），胃癌组织为阴性对照（上皮细胞无着色）。
