The encoded protein is involved in cell-to-cell interactions as well as cell-matrix interactions during development and differentiation. The encoded protein has been shown to be involved in development of the nervous system, and for cells involved in the expansion of T cells and dendritic cells which play an important role in immune surveillance. 靶点与功能

CD56（神经细胞黏附分子，NCAM）是Ig超家族跨膜糖蛋白，分子量120-220kDa（因剪接异构体差异），像一把“多功能钥匙”：

免疫领域：主要表达于NK细胞、NK-T细胞、部分活化T细胞，是NK细胞分选和功能研究的“金标准”标志物；

神经领域：参与神经细胞黏附、突触形成和神经再生，在阿尔茨海默病、神经损伤修复研究中常用；

肿瘤领域：神经内分泌肿瘤（如小细胞肺癌）、NK细胞淋巴瘤中高表达，可辅助病理诊断。

实验操作关键要点

流式细胞术（NK细胞检测）：

样本处理：外周血单个核细胞（PBMC）用PBS+2%FBS洗涤2次，调整浓度至1×10⁶/mL；

染色方案：PE标记CD56抗体（如bsm-30085M-PE）与APC-CD3抗体（1:100）避光孵育20分钟（4℃），红细胞裂解后上机；

设门策略：CD3⁻CD56⁺为NK细胞群，CD3⁺CD56⁺为NKT细胞群（正常占比约5%-15%）。

IHC-P（神经内分泌肿瘤切片）：

抗原修复：柠檬酸钠缓冲液（pH6.0）微波修复15分钟（95℃），冷却后用3% H₂O₂阻断内源性过氧化物酶；

阳性定位：肿瘤细胞胞膜/胞质呈棕黄色颗粒状阳性，神经内分泌肿瘤（如小细胞肺癌）阳性率＞90%；

对照设置：用扁桃体组织作阳性对照（NK细胞显色），胃癌组织作阴性对照（上皮细胞不表达）。

WB验证（SK-N-SH神经母细胞瘤细胞）：

样本制备：RIPA裂解液（含蛋白酶抑制剂）冰上裂解30分钟，12000g离心取上清，上样量50μg总蛋白；

电泳条件：8% SDS-PAGE凝胶（分离高分子量蛋白），转膜用0.45μm PVDF膜，5% BSA封闭1小时；

抗体孵育：一抗1:1000稀释（4℃过夜），HRP二抗（山羊抗小鼠）1:5000室温孵育1小时，曝光可见180-220kDa条带（异构体混合）。
