产品介绍Cytolytic T lymphocytes (CTL) and natural killer (NK) cells share the remarkable ability to recognize, bind, and lyse specific target cells. They are thought to protect their host by lysing cells bearing on their surface 'nonself' antigens, usually peptides or proteins resulting from infection by intracellular pathogens. The protein encoded by this gene is crucial for the rapid induction of target cell apoptosis by CTL in cell-mediated immune response. [provided by RefSeq, Jul 2008]

靶点与功能

颗粒酶B（Granzyme B, GZMB）是丝氨酸蛋白酶家族成员，由细胞毒性T淋巴细胞（CTL）和自然杀伤（NK）细胞分泌，通过切割胱天蛋白酶前体（如procaspase-3） 诱导靶细胞凋亡，同时参与细胞因子加工和细胞外基质降解，在抗肿瘤免疫、自身免疫疾病及炎症反应中起核心作用。其异常表达与肿瘤免疫逃逸、病毒感染及移植排斥反应密切相关，是评估免疫细胞杀伤活性的重要标志物。

实验操作关键要点

WB检测（CTL/NK细胞裂解液）：

样本处理：使用含1mM PMSF和10μM E-64的RIPA裂解液（抑制丝氨酸蛋白酶活性），冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜选用0.45μm PVDF膜（250mA恒流1.5小时）；

抗体孵育：一抗1:1000稀释（5% BSA+TBST），4℃孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间30-60秒。

IHC-P组织染色（肿瘤微环境样本）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），冷却至室温后用3% H₂O₂阻断内源性过氧化物酶10分钟；

阳性定位：CTL/NK细胞胞质呈棕黄色颗粒状阳性，在肿瘤浸润淋巴细胞（TILs）中高表达；

对照设置：必须包含NK细胞系（如YTS） 作为阳性对照，CD8⁺ T细胞敲除组织作为阴性对照。

IF双标实验（免疫荧光共定位）：

荧光标记：一抗与CD8抗体（小鼠单抗）共孵育，二抗选用Alexa Fluor 488（抗兔）和Cy3（抗小鼠），4℃避光孵育2小时；

封片处理：使用含DAPI的抗淬灭封片剂，激光共聚焦显微镜下观察（激发波长488nm/550nm）。
