They are thought to protect their host by lysing cells bearing on their surface 'nonself' antigens, usually peptides or proteins resulting from infection by intracellular pathogens. 颗粒酶B(Granzyme B, GZMB)是丝氨酸蛋白酶家族成员,由细胞毒性T淋巴细胞(CTL)和自然杀伤(NK)细胞分泌,通过切割胱天蛋白酶前体(如procaspase-3) 诱导靶细胞凋亡,同时参与细胞因子加工和细胞外基质降解,在抗肿瘤免疫、自身免疫疾病及炎症反应中起核心作用。其异常表达与肿瘤免疫逃逸、病毒感染及移植排斥反应密切相关,是评估免疫细胞杀伤活性的重要标志物。 实验操作关键要点
WB检测(CTL/NK细胞裂解液): 样本处理:使用含1mM PMSF和10μM E-64的RIPA裂解液(抑制丝氨酸蛋白酶活性),冰上裂解30分钟,12000g离心15分钟取上清; 电泳条件:12% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,转膜选用0.45μm PVDF膜(250mA恒流1.5小时); 抗体孵育:一抗1:1000稀释(5% BSA+TBST),4℃孵育过夜,HRP标记二抗(山羊抗兔)1:5000室温孵育1小时,曝光时间30-60秒。 IHC-P组织染色(肿瘤微环境样本): 抗原修复:采用柠檬酸钠缓冲液(pH6.0) 微波修复20分钟(95℃),冷却至室温后用3% H₂O₂阻断内源性过氧化物酶10分钟; 阳性定位:CTL/NK细胞胞质呈棕黄色颗粒状阳性,在肿瘤浸润淋巴细胞(TILs)中高表达; 对照设置:必须包含NK细胞系(如YTS) 作为阳性对照,CD8⁺ T细胞敲除组织作为阴性对照。 IF双标实验(免疫荧光共定位): 荧光标记:一抗与CD8抗体(小鼠单抗)共孵育,二抗选用Alexa Fluor 488(抗兔)和Cy3(抗小鼠),4℃避光孵育2小时; 封片处理:使用含DAPI的抗淬灭封片剂,激光共聚焦显微镜下观察(激发波长488nm/550nm)。 公司正在出售的产品
