产品介绍Glutathione S-transferases (GSTs) are a family of enzymes that play an important role in detoxification by catalyzing the conjugation of many hydrophobic and electrophilic compounds with reduced glutathione. Based on their biochemical, immunologic, and structural properties, the soluble GSTs are categorized into 4 main classes: alpha, mu, pi, and theta. This GST family member is a polymorphic gene encoding active, functionally different GSTP1 variant proteins that are thought to function in xenobiotic metabolism and play a role in susceptibility to cancer, and other diseases. 靶点与功能

GSTP1（谷胱甘肽硫转移酶pi）是II相解毒酶家族成员（分子量约23kDa），通过催化谷胱甘肽（GSH）与亲电物质结合，参与药物代谢、氧化应激解毒及肿瘤耐药过程。其核心研究价值：

肿瘤耐药机制：在肺癌、乳腺癌中高表达，通过代谢化疗药物（如顺铂、阿霉素）导致耐药，是逆转耐药的潜在靶点；

氧化应激：清除活性氧（ROS），保护细胞免受氧化损伤，与神经退行性疾病（如阿尔茨海默病）相关；

毒理学研究：作为环境污染物（如重金属、多环芳烃）暴露的生物标志物。

实验操作关键要点

WB检测（肿瘤细胞样本）：

样本处理：使用含1mM DTT的RIPA裂解液（维持酶活性）冰上裂解30分钟，12000g离心15分钟取上清；

电泳条件：12% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，转膜用0.45μm PVDF膜（250mA恒流1小时）；

抗体孵育：一抗推荐1:1000稀释（5%脱脂奶粉+TBST），4℃摇床孵育过夜，HRP二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P组织染色（肺癌切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复15分钟（95℃），冷却至室温后PBS洗3次；

阳性对照：推荐使用肺腺癌组织（癌细胞胞质呈棕黄色颗粒状阳性）；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，降低非特异性结合。
