产品介绍This gene encodes a member of the CD1 family of transmembrane glycoproteins, which are structurally related to the major histocompatibility complex (MHC) proteins and form heterodimers with beta-2-microglobulin. The CD1 proteins mediate the presentation of primarily lipid and glycolipid antigens of self or microbial origin to T cells. The human genome contains five CD1 family genes organized in a cluster on chromosome 1. The CD1 family members are thought to differ in their cellular localization and specificity for particular lipid ligands. The protein encoded by this gene localizes to the plasma membrane and to recycling vesicles of the early endocytic system. Alternative splicing results in multiple transcript variants. 靶点与功能

CD1分子是非多态性MHC-I类样跨膜糖蛋白，家族包括CD1a、CD1b、CD1c、CD1d和CD1e五个成员，主要表达于抗原呈递细胞（APC）及胸腺皮质细胞表面。其核心功能是通过抗原结合槽捕获脂质和糖脂类抗原（如分枝杆菌脂阿拉伯甘露聚糖、肿瘤糖脂），形成CD1-抗原复合体后提呈给NKT细胞、γδ T细胞等，激活天然免疫和适应性免疫应答，参与抗感染、抗肿瘤及自身免疫调节。

实验操作关键要点

WB优化（APC细胞样本）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；推荐使用树突状细胞（DC） 作为阳性对照（CD1高表达）；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保34kDa主条带与杂带分离；

抗体孵育：一抗推荐1:500稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项（胸腺组织切片）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强膜蛋白信号；

阳性定位：胸腺皮质细胞、朗格汉斯细胞及树突状细胞表面呈棕黄色阳性；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，降低非特异性结合。

Flow-Cyt检测要点（外周血单核细胞）：

样本制备：EDTA抗凝全血经Ficoll密度梯度离心分离PBMC，PBS洗涤后用2% FBS重悬；

抗体孵育：1μg抗体/1×10⁶细胞，4℃避光孵育30分钟，流式缓冲液洗涤2次后上机；

荧光标记：推荐与CD11c-PE（DC标志物）共染，通过流式细胞术分析CD1⁺CD11c⁺细胞比例。
