靶点与功能

CR2（Complement Receptor 2，CD21）是表达于B细胞、滤泡树突状细胞（FDC） 表面的跨膜糖蛋白，通过结合补体片段C3d及EB病毒gp350蛋白，参与B细胞活化共刺激信号传递（与CD19/CD81形成共受体复合物）、免疫复合物捕获及抗原呈递。其功能异常与系统性红斑狼疮（SLE）、B细胞淋巴瘤及EB病毒相关疾病密切相关，是研究B细胞免疫调控的关键分子标志物。

实验操作关键要点

WB优化（B细胞样本）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；推荐使用Raji细胞系（Burkitt淋巴瘤细胞，CR2高表达）作为阳性对照；

电泳条件：7.5% SDS-PAGE凝胶（因分子量较大），恒压80V浓缩胶、120V分离胶，电泳时间90分钟确保条带分离；

抗体孵育：一抗推荐1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时，曝光时间控制在60-90秒。

FC（流式细胞术）注意事项：

样本制备：全血经红细胞裂解液处理后，PBS洗涤2次，调整细胞浓度至1×10⁶/mL；

抗体孵育：推荐1:100稀释抗体（含2% FBS的PBS），4℃避光孵育30分钟，洗涤后用流式细胞仪检测（激发波长488nm，发射波长525nm）；

阳性对照：正常外周血B细胞（CD19⁺CD21⁺） 呈阳性信号，可通过CD19/CD21双标验证细胞纯度。

IF双标实验（B细胞活化研究）：

推荐与CD19抗体共染，使用Alexa Fluor 488（绿色，标记CR2）和594（红色，标记CD19），激光共聚焦观察B细胞表面共受体复合物的共定位。
