It has essential roles in B cell growth and differentiation, and the regulation of IgE production. This protein also exists as a soluble secreted form, then functioning as a potent mitogenic growth factor. 靶点与功能

CD23（FcεRII）是一种C型凝集素家族成员，作为IgE低亲和力受体，主要表达于B细胞、单核细胞及树突状细胞表面，通过与IgE结合参与B细胞增殖分化、IgE合成调控及抗原呈递过程。其异常表达与过敏性疾病（如哮喘）、B细胞淋巴瘤及自身免疫病密切相关，是研究免疫失衡机制的重要分子标志物。

实验操作关键要点

WB优化（B细胞样本）：

样本处理：使用含蛋白酶抑制剂（1mM PMSF） 的RIPA裂解液冰上裂解30分钟，12000g离心15分钟取上清；推荐使用Raji细胞系（Burkitt淋巴瘤细胞，高表达CD23）作为阳性对照；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，电泳时间90分钟确保45kDa条带分离；

抗体孵育：一抗推荐1:1000稀释（5%脱脂奶粉+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项（扁桃体组织）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强细胞膜信号；

阳性定位：正常扁桃体组织中生发中心B细胞胞膜呈棕黄色阳性，淋巴瘤组织可见弥漫性强阳性；

封闭处理：5%驴血清+0.3% Triton X-100室温封闭45分钟，减少非特异性结合。

FC检测要点（外周血B细胞）：

样本制备：EDTA抗凝全血经红细胞裂解液处理后，PBS洗涤2次，调整细胞浓度至1×10⁶ cells/mL；

抗体孵育：CD23抗体按1:200稀释，4℃避光孵育30分钟，流式缓冲液洗涤后上样检测，推荐与CD19抗体联合标记（双阳性为B细胞群体）。
