Ca2+/calmodulin-dependent kinase II γ (CaMKIIγ) gene is mapped to human chromosome 10q22.2. The encoded protein is a multifunctional serine/threonine kinase and is specific to various substrates. CaMKII contains four isoforms: CaMKIIα/β/γ/δ. CaMKII is ubiquitously expressed in humans. The protein consists of an N-terminal catalytic domain, a regulatory domain, and a C-terminal association domain.
CaM-kinase II (CAMK2) is a prominent Ser/Thr protein kinase in the central nervous system that may function in long-term potentiation and neurotransmitter release. Likely autophosphorylation of Thr-286 allows the kinase to switch from a calmodulin-dependent to a calmodulin-independent state. CAMK2 is composed of four different chains: alpha, beta, gamma, and delta. The different isoforms assemble into homo- or heteromultimeric holoenzymes composed of 8 to 12 subunits.

CaMKIIγ participates in the β-catenin, Stat3 (signal transducer and activator of transcription 3), and NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) signal pathways..
