Members of this Type 1 transmembrane protein family share structural characteristics including an extracellular domain consisting of multiple epidermal growth factor-like (EGF) repeats, and an intracellular domain consisting of multiple, different domain types. Notch family members play a role in a variety of developmental processes by controlling cell fate decisions. The Notch signaling network is an evolutionarily conserved intercellular signaling pathway which regulates interactions between physically adjacent cells. In Drosophilia, notch interaction with its cell-bound ligands (delta, serrate) establishes an intercellular signaling pathway that plays a key role in development. Homologues of the notch-ligands have also been identified in human, but precise interactions between these ligands and the human notch homologues remain to be determined. This protein is cleaved in the trans-Golgi network, and presented on the cell surface as a heterodimer. IHC-P肿瘤组织染色(小细胞肺癌样本): 抗原修复:柠檬酸盐缓冲液(pH6.0)微波修复15分钟(石蜡切片),胞质信号显著增强; 阳性定位:肿瘤细胞胞质呈棕黄色颗粒状阳性,小细胞肺癌组织阳性率>90%,腺癌/鳞癌阴性(可与TTF-1联合检测提高特异性); 对照设置:正常脑组织(神经元胞质阳性对照)和肺腺癌组织(阴性对照)。 WB验证(神经母细胞瘤细胞系): 样本制备:SK-N-SH细胞用含蛋白酶抑制剂(1mM PMSF) 的RIPA裂解液冰上裂解30分钟,12000g离心取上清; 电泳条件:10% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,转膜使用0.45μm PVDF膜(250mA恒流1.5小时); 抗体孵育:一抗1:1000稀释(5% BSA+TBST),4℃孵育过夜,HRP二抗(山羊抗兔)1:5000室温孵育1小时,曝光时间45秒(90kDa处可见清晰二聚体条带)。 ELISA血清定量检测: 包被优化:抗体用碳酸盐缓冲液(pH9.6)稀释至2μg/mL,4℃包被过夜; 样本处理:血清需避免溶血(红细胞含NSE会干扰结果),1:10稀释后37℃孵育1.5小时; 显色控制:TMB底物显色10分钟,2M H₂SO₄终止,450nm读取OD值(标准曲线范围0.1-100ng/mL)。 公司正在出售的产品
