靶点与功能

滑膜细胞凋亡抑制物1（SYVN1，又称Synoviolin）是一种E3泛素连接酶，通过介导错误折叠蛋白的泛素化降解参与内质网应激反应，在性关节炎、滑膜细胞过度增殖、抗凋亡信号通路激活中发挥核心作用。其功能异常会导致滑膜组织炎症迁延和关节破坏，是自身免疫性疾病机制研究的重要靶点。

实验操作关键要点

WB优化（滑膜组织样本）：

样本处理：滑膜组织需用含1% Triton X-100和蛋白酶抑制剂的RIPA裂解液冰上裂解40分钟，12000g离心15分钟取上清；

电泳条件：10% SDS-PAGE凝胶，恒压80V浓缩胶、120V分离胶，确保67kDa主条带与杂带分离；

抗体孵育：一抗推荐1:1000稀释（5% BSA+TBST），4℃摇床孵育过夜，HRP标记二抗（山羊抗兔）1:5000室温孵育1小时。

IHC-P注意事项（关节炎滑膜组织）：

抗原修复：采用柠檬酸钠缓冲液（pH6.0） 微波修复20分钟（95℃），增强胞质信号；

阳性对照：推荐使用关节炎患者滑膜组织切片（滑膜衬里层细胞胞质呈棕黄色阳性）；

封闭处理：5%驴血清室温封闭30分钟，配合3% H₂O₂阻断内源性过氧化物酶（10分钟）。
