商品属性 产品介绍The protein encoded by this gene belongs to the family of P-type cation transport ATPases, and to the subfamily of Na+/K+-ATPases. Na+/K+ -ATPase is an integral membrane protein responsible for establishing and maintaining the electrochemical gradients of Na and K ions across the plasma membrane. These gradients are essential for osmoregulation, for sodium-coupled transport of a variety of organic and inorganic molecules, and for electrical excitability of nerve and muscle. This enzyme is composed of two subunits, a large catalytic subunit (alpha) and a smaller glycoprotein subunit (beta). The catalytic subunit of Na+/K+ -ATPase is encoded by multiple genes. This gene encodes an alpha 1 subunit. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, May2009].

PubMedPubMed 靶点与功能 转录因子SOX10(SRY-related HMG-box gene 10)是SOX家族C亚族成员,通过结合靶基因启动子的HMG结构域调控转录,在神经嵴细胞分化、周围神经系统发育及黑色素细胞生成中起核心作用。其功能异常与神经嵴来源肿瘤(如黑色素瘤、神经鞘瘤、雪旺细胞瘤)密切相关,是这类肿瘤病理诊断的特异性标志物,也参与肌上皮肿瘤的鉴别诊断。 实验操作关键要点 IHC-P优化(黑色素瘤病理诊断): 样本处理:福尔马林固定石蜡包埋组织需用pH9.0 EDTA缓冲液热诱导抗原修复(95℃,20分钟),充分暴露核内抗原表位; 抗体孵育:一抗推荐1:200稀释(5%驴血清+TBST),4℃湿盒孵育过夜,HRP标记二抗(山羊抗兔)1:500室温孵育30分钟; 显色控制:DAB显色时间3-5分钟,阳性信号定位于肿瘤细胞核(呈棕黄色颗粒状),阴性对照(正常皮肤组织)仅表皮基底层少量黑色素细胞阳性。 WB检测要点(神经嵴细胞样本): 样本处理:使用含蛋白酶抑制剂(1mM PMSF) 的RIPA裂解液冰上裂解30分钟,12000g离心15分钟取上清; 电泳条件:10% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,确保47kDa主条带与杂带分离; 阳性对照:推荐使用SK-MEL-28黑色素瘤细胞裂解液(47kDa条带清晰可见)。 IF双标实验(细胞定位研究): 推荐与S100蛋白共染,使用Alexa Fluor 488(绿色,标记SOX10,核定位)和594(红色,标记S100,胞质定位),激光共聚焦观察神经嵴细胞共表达模式。 公司正在出售的产品
