PubMedPubMed 产品介绍The protein encoded by this gene is one of the highly conserved mini-chromosome maintenance proteins (MCM) that are essential for the initiation of eukaryotic genome replication. The hexameric protein complex formed by the MCM proteins is a key component of the pre-replication complex (pre_RC) and may be involved in the formation of replication forks and in the recruitment of other DNA replication related proteins. The MCM complex consisting of this protein and MCM2, 4 and 6 proteins possesses DNA helicase activity, and may act as a DNA unwinding enzyme. Cyclin D1-dependent kinase, CDK4, is found to associate with this protein, and may regulate the binding of this protein with the tumorsuppressor protein RB1/RB. IHC-P肿瘤增殖检测(石蜡切片): 抗原修复:采用柠檬酸盐缓冲液(pH6.0) 微波修复20分钟(95℃),增强核信号; 阳性定位:肿瘤组织中增殖区细胞胞核呈棕黄色阳性,正常组织中仅见于基底层或生发中心; 对照设置:需包含结肠腺癌组织(高增殖阳性对照)和正常肝组织(低增殖阴性对照)。 WB验证(细胞裂解液): 样本处理:HeLa细胞(对数生长期)用含1mM PMSF的RIPA裂解液冰上裂解30分钟,12000g离心取上清; 电泳条件:10% SDS-PAGE凝胶,恒压80V浓缩胶、120V分离胶,转膜使用0.45μm PVDF膜(250mA恒流1.5小时); 抗体孵育:一抗1:1000稀释(5%脱脂奶粉+TBST),4℃孵育过夜,HRP二抗(山羊抗兔)1:5000室温孵育1小时,曝光30-60秒(目标条带约82kDa)。 公司正在出售的产品
