PubMedPubMed 产品介绍This gene encodes a classical cadherin of the cadherin superfamily. Alternative splicing results in multiple transcript variants, at least one of which encodes a preproprotein that is proteolytically processed to generate the mature glycoprotein. This calcium-dependent cell-cell adhesion protein is comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Mutations in this gene are correlated with gastric, breast, colorectal, thyroid and ovarian cancer. Loss of function of this gene is thought to contribute to cancer progression by increasing proliferation, invasion, and/or metastasis. The ectodomain of this protein mediates bacterial adhesion to mammalian cells and the cytoplasmic domain is required for internalization. IHC-P优化(结直肠癌组织): 抗原修复:采用柠檬酸钠缓冲液(pH6.0) 微波修复20分钟(95℃),增强膜/浆信号; 阳性定位:正常上皮细胞细胞膜/胞质呈连续棕黄色阳性,肿瘤组织中因EMT呈阴性或细胞膜信号断裂; 对照设置:必须包含正常结肠黏膜(强阳性)和间质纤维组织(阴性对照)。 WB检测注意事项: 样本处理:使用含1mM PMSF的RIPA裂解液冰上裂解30分钟,12000g离心取上清(避免蛋白酶降解); 电泳条件:8% SDS-PAGE凝胶(因分子量较大),恒压80V浓缩胶、100V分离胶,目标条带约120kDa; 抗体孵育:一抗1:1000稀释(5% BSA+TBST),4℃摇床孵育过夜,HRP二抗(山羊抗兔)1:5000室温孵育1小时。 公司正在出售的产品
