PubMedPubMed 产品介绍Cytochrome C is an electron transporting protein that resides within the intermembrane space of the mitochondria, where it plays a critical role in the process of oxidative phosphorylation and production of cellular ATP. An increasing amount of interest has been directed toward the role which cytocrome C has been demonstrated to play in apoptotic processes. Following exposure to apoptotic stimuli, cytochrome C is rapidly released from the mitochondria into the cytosol, an event which may be required for the completion of apoptosis in some systems. Cytosolic cytochrome C functions in the activation of caspase 3, an ICE family molecule that is a key effector of apoptosis. 靶点与功能
细胞色素C(Cytochrome C, Cyt C)是线粒体电子传递链的关键蛋白,位于线粒体内膜间隙,通过血红素铁离子(Fe²⁺/Fe³⁺)的氧化还原传递电子,参与ATP生成。当细胞受到凋亡刺激(如DNA损伤、氧化应激)时,Cyt C会从线粒体释放至胞质,与Apaf-1结合形成“凋亡小体”,激活caspase-9,最终启动 caspase级联反应导致细胞凋亡。其释放是线粒体凋亡通路的“分子开关”,也是区分凋亡与坏死的重要标志物。 实验操作关键要点
WB检测(凋亡细胞模型): 样本制备:使用线粒体/胞质分离试剂盒分离胞质组分,避免线粒体破裂导致假阳性; 电泳条件:15% SDS-PAGE凝胶(因分子量小),恒压120V电泳60分钟,转膜选用0.22μm PVDF膜; 抗体孵育:兔单抗推荐1:2000稀释(5%脱脂奶粉+TBST),4℃孵育过夜,曝光时间10-30秒(避免信号过强)。 IF凋亡定位(Hela细胞): 染色步骤:4%多聚甲醛固定15分钟,0.2% Triton X-100透化10分钟,5%驴血清封闭30分钟; 共染策略:Cyt C抗体(绿色荧光)与线粒体 marker(Tom20,红色荧光)共染,正常细胞呈“红绿共定位”(黄色),凋亡细胞绿色信号弥散至胞质; 激发波长:FITC通道(488nm)检测Cyt C,Cy3通道(555nm)检测线粒体。 IHC-P组织染色(脑缺血模型): 抗原修复:采用柠檬酸钠缓冲液(pH6.0) 微波修复15分钟(95℃),冷却至室温; 阳性定位:正常组织中线粒体呈颗粒状阳性(心肌、脑组织强表达),凋亡区域胞质弥漫性着色; 对照设置:必须包含正常组织(线粒体阳性)和凋亡诱导组织(胞质阳性)作为对照。 公司正在出售的产品
